not recommended. TEMED induces the radicalization of APS to crosslink Bisacrylamide and acrylamide. Therefore, you want this reaction to take place as quickly and efficiently as possible. It only takes about 3-4 minutes to mix all the reagents together and 45-60 minutes for polymerization. If you want to expedite things and know you'll be needing a few gels in the upcoming days, simply pour a few plates the day or two before, let them polymerize and wrap the glass plates (layered in wet paper towels) in saran wrap and store them at 4C with the combs in-place! This will prevent the gels from shrinking and can be stored (with success) up to 4 days prior to use.

It is possible to mix all the ingredients including APS and TEMED and make the gel for SDS-PAGE which can be store at 4 C for 1 or maximum 2 days but not more than that for best result.

It's better to mix acrylamide, buffer, SDS, water, APS and TEMED before each gel electrophoresis. But the gels which are left can be sealed in zip lock bags with a little DD water and preseverd at 4 oC for a few days. Don't mix the TEMED and APS together. APS can be frozened at -20 oC in 1.5 ml tubes, avoid thawing and refreezing. TEMED can be preserved in room tempreture.

In our lab we usually use premixed solution without APS and TEMED. It's stable for a few month at +4. We also keep ready plates wrapped in wet paper in zip lock bags at +4 for a day or two, and froze 10% APS in 1,5ml tubes.

Just store it without the APS and the TEMED and I think you should be fine. Otherwise, prepare it fresh, which I personally prefer.

Storage will cause serious problem in pH. pH alteration will give unexpected protein profile. Then again SDS precipitation you can see while storing in cold condition. These are the problems what I know. To reconstitute hardly it will take 5 minutes. So, I suggest that prepare fresh.

I have found that I can pour the resolving gel and let it completly polymerize. Then it may be stored immersed in resolving gel buffer at 4 degrees and it is good for up to a week. I always pour the stacker just before use however. This has given me good results.

There is no need to add SDS to the gel if it is in the sample and running buffers. All gel manufacturers stopped adding it several years ago as it destabilizes the gels during storage and running. Biorad has some documentation on this, or see Novex Bistris documents.

Your premix without SDS should be stable for months sans gel polymerization agents. Bring it to room temperature before polymerizing as Tris pH is temperature dependent.

Not at all recommended. You have to prepare a fresh every time which takes few minutes and of course you have to wait for polymerization step. Gel quality will be not good.

The gels can be prepared previous evening and can be stored in fridge for "Maturation". Keep the upper and lower edges moist by keeping them in contact with water.

I have had the best results after storing the gels overnight..............

Alternatively you can use Gel Storage buffer on the top of the gels  in order to preserve the moisture of gels.
