Mass spec is showing 8Da more for a protein, what could be the reason? LCMS was carried without deglyco and with out reducing the protein ?
The mass spec may be miscalibrated. If you have not done so, perform a calibration just before running your sample.

The subsitiution of a Glu residue with a His residue will lead to a mass increase of 8.016319Da.
Is this a possibility for your work?
