I need a help from you people . Actually I am trying to develop KLN205 renal cancer syngenic tumor model .
I have used both Balb/c mice and C57BL/6 mice and 1 and 2 million of KLN205 cell were implanted with Matrigel(1:1) . After a week a palpable tumor were seen but they regressed
Could please help me to solve my problem about this model
We've done several kidney xenografts commercially (such as for 786-O: http://altogenlabs.com/xenograft-models/kidney-cancer-xenograft/786o-xenograft-model/) and usually the issues with tumor regression are caused by the effects of a functioning immune system. Athymic Balb/c mice will generally suffice, but NOD/SCID mice are generally the safest bet for these kinds of studies.
You may use cyclosporine few days before the inoculation of cells and continue till 1 week. But as you start dosing of the animals, cyclosporine must be stopped to see the effect of the treatment.
Tumor regression is nothing unusual! I have worked on LA-7 induced syngeneic mammary tumor model (in SD rats) and I have also faced such issues. However, application of immunosuppressant (cyclosporine) results in better development of tumor in rats (with reduced regression rate). Refer literature for the dosing regimen of immunosuppressant in mice, as it has severe side effects if overdosed. It must be ensured that application of immunosuppressant is discontinued before onset of treatment or else it may affect the therapeutic efficacy of drug under investigation.
