Shouqiang Cheng , You can consider the Zymo Research YeaStar Genomic DNA Kit or the Qiagen Yeast DNA Extraction Kit for total DNA isolation from yeast. If plasmid recovery is also a priority, the Zymo Research Zymoprep Yeast Plasmid Miniprep II or the Promega Wizard Genomic DNA Purification Kit may be useful. MP Biomedicals FastDNA SPIN Kit is another solid option, especially for tough yeast cell walls. Thank you!
Thanks for your response! I want to determine the copy number of a plasmid in yeast cells so I need isolate both genomic and plasmid DNA from the cell. I did not find a good commercially available kit for the experiment.
Most of kits are column based, so they might not solve your purpose. I guess below mentioned kit do total lysis and don't require comlumn for genomic dna purification. So this might be useful for you. Use chloroform cleaning for your genomic DNA, present in soup at protein precipitation step. You can repeat chloroform cleaning step to get highest quality genomic dna along with plasmid.
This is the protocol I used for genomic DNA extraction from Pichia Pastoris. This should extract both genomic and plasmid DNA, however, plasmid DNA will be low. I used this method for my Pichia transformants.
Protocol for DNA Extraction from Pichia Transformants
Collect a single colony of Pichia transformant from a 100 µl overnight culture in YPD medium and pellet the cells by centrifugation.
Resuspend the cell pellet in 50 µl of Cellytic Y (Sigma) containing 5 mM DTT.
Incubate the mixture at 65°C for 10 minutes, then allow it to cool.
Add 60 µl of Phenol:Chloroform:Isoamyl Alcohol (25:24:1) and mix thoroughly.
Centrifuge at room temperature at 13,000 rpm for 20 minutes.
Transfer the aqueous phase to a fresh microfuge tube.
Add 1/10 volume of 3M sodium acetate and 2.5 volumes of 100% ethanol to precipitate the DNA.
Incubate the mixture at -20°C overnight.
Centrifuge at 13,000 rpm for 2 minutes at 4°C to pellet the DNA.
Wash the DNA pellet twice with 200 µl of ice-cold 70% ethanol, centrifuging for 1 minute each time.
Air-dry the pellet at 37°C before resuspending in an appropriate buffer for downstream applications. I dissolved in 50 µl deionised water (10 min/50 ºC).
