Please correct me if I am wrong- Total RNA extracted from eukaryotic cells has three distinct visible bands on an agarose gel- 28SrRNA, 18SrRNA and the least prominent 5SrRNA & 5.8SrRNA. And 28SrRNA is more abundant than 18SrRNA in the normal eukar cell, thus showing greater band intensity in agarose gel. For normal eukaryotic cells the intensity of 28SrRNA is twice that of 18SrRNA but in picture attached (i.e, RNA extracted from tumor cells) 28SrRNA is fainter than 18SrRNA. Kindly explain this aberration
not 100% percent sure, but this is a sign of poor quality of isolated RNA (hydrolysis)
Pay attention to all plasticware you are using, make sure they are RNAase free, all glass containers should be dry-sterilized in high temperature. Also pay attention to solutions used during RNA isolation and the storage buffer
