While I haven't worked with streptococcus species, I have experience purifying proteins from other Gram positive species. The easiest way to prepare the bacteria for lysis and protein purification is to make them into protoplasts first and sonicate them. A protocol for making streptococcus protoplasts can be found in the paper below. Briefly, grow the cells in a medium that is supplemented with glycine, and remove the cell wall with lysozyme. This will give you fragile cells that you can lyse and use in whichever protein purification strategy you want.
