I want to isolate the brightest bioluminescent mutants of photobacterium phosphoreum from my culture, but all colonies look equally as bright. Any ideas, how I could isolate brighter mutants??

Although I don't want to do gene engineering (at least not with P. kishitanii), I think I will try the mutant selection as described
Yes, try mutant selection. Also there are some amino acids like Arginine, which when added to the cultivation medium, will bring out the brightest bioluminescence. See the prescribed incubation conditions, related to time and temperature.
Flow cytometer with cell sorting can work too. Expensive machine but you can try a parternship with universities
