I did some in vivo imaging using a V5 epitope-tagged protein in C. elegans germline. V5 tag works really well for Immunohistochemistry and generally gives less background. But now I am looking forward to doing some biochemical analyses and intend to do Mass spec to identify protein partners of my protein of interest. I am curious to know if the V5 tag would be a good one for biochemical/Mass spec analyses as well. I came to know FLAG peptide is a good tag for biochemistry. But in order to use FLAG tag, I need to generate new alleles which albeit not hard are time-consuming. I need some quick and dirty experiments to get a preliminary idea if Mass spec works or not and then expand my project. Does anyone have any suggestions/insights? Also if anyone has any idea how to elute V5 tag protein to prepare the sample for Mass Spec analysis, please share.
