I've conducted some checkerboard assays to determine the fractional inhibitory concentration on several Candida sp. However, I've been getting some odd readings particularly at the higher concentrations. Kindly find a copy of my results as attached.
The results are presented as percentage of cell viability. The MIC for the fluconazole and chitosan on their own are 0.781 ug/mL and 3.91 ug/mL, respectively. Please note that the fluconazole concentration decreases from right-to-left whereas, chitosan concentration decreases from top-to-bottom. The observation I've mentioned can be in seen in the top rows and columns towards the right. The percentage of cell viability at higher concentration of fluconazole-chitosan seems to be higher than those treated with lower concentration. I've observed the same occurrence in the replicates as well as in independent assays.
Is this indicative of some form of antagonism? Aside from that, is there a way to overcome this as I'm not quite sure how to justify this since there is no apparent trend moving from a lower drug combination to a higher one?
