I've been growing a primary cell line for 2 months now and in the last month growth has slowed down significantly and more cells have become senescent. Since subculturing the cells seemed to have revived and growth is a lot better but mycoplasma results from a DAPI stain seem a bit concerning.

According to some reference on internet : Mycoplasma staining with DAPI appears as a fine particulate or filamentous staining over the cytoplasm at 100X magnification. Nuclei of the cells are also brightly stained by this method and thereby act as endogenous positive control for the staining procedure.... individual mycoplasma are about 0.3 um....
