The enclosed figure is an fEPSP trace I recorded from rat motor cortex, with the recording electrode placed at layer II/III and the stimulating electrode at layer V (stimulating the vertical afferent fiber coming from white matter).
I always thought that with a negative-going fEPSP curve, the inverted trough preceding the negative-going fEPSP reflects a presynaptic fiber volley. Recently I read an article (Dasari and Yuan, Toxicol Appl Pharmacol, 2009, 240: 412-422; done in visual cortex with identical stimulation and recording arrangement) saying that this is a population spike caused by antidromic stimulation of the axon. I believe that my stimulation method should be orthodromic. How did I achieve antidromic stimulation? So I am confused. Which one is it ? a fiber volley or population spike? Hope someone can give me an answer.
I would like to know why and how to tell the differences. Thanks in advance!
