You are probably concerned that Nanog was not included in the 4 factors when listed as indispensable in the last sentence. Nanog is not indispensable if the other 4 are present, but can replace c-Myc or Klf4, hence the Sox2 ,Oct4 and Nanog. Poorly worded hence leading to confusion. In cancer stem cells (CSCs), Nanog is supposed to be specific for epithelial tumor CSCs, while Oct4 is present in both mesenchymal and epithelial tumor CSCs.
Christopher, how good is the data on Oct4 in CSCs? There is a lot of bad data on Oct4 in adult stem cells; I am less familiar with the CSC field. When we first did the OCt4 expression with Hans Schoeler in 1990, Oct4 was only in the germline and in pregastrula embryos. Jaenisch has shown Oct4 is dispensable for all adult stem cells.
One of the fascinating aspects of cancer is that most all cancers rarely if ever escape their cell lineages. This would suggest that there are fixed epigentic imprints on chromatin, which are likely set in development, are are not erased in the CSC. If Oct4 was acting in CSCs you might expect at least some to achieve totipotency. Any insights?
