Generally in biomedical science, measurement of "cortisol" is used, but I am looking for some "pen & paper" procedure that strikes out blood sampling or anything like that.
I am a little unsure what exactly you mean by "during a particular task" or what kind of task you mean by this. But it might be worthwhile to have a look at the following German inventory, which was translated to English and validated:

If you search e.g. "allostatic load", you will see that many other biomarkers than cortisol are used to measure stress (and none are gold standards).
If you search "stress questionnaire" you will see many different instruments.
If you give a little more information on your study plans, it will be easier to recommend one.
