Hi there! I am wondering if there is any way of transiently increase platelet count in mice, like administrating TPO or something like that?. Thanks a lot!
subcutaneous injection of purified recombinant HST-1 protein into the animals for two consecutive days. An increase in the number of platelets was observed with a peak at day 10, followed by a quick decrease to normal levels
@Heyder the reference you provided is a protocol for adenovirus expressing HST-1.
Is there any commercial supplier that provides purified HST-1?
Plus the paper said "The HST-1 (fibroblast growth factor 4, FGF4) protein is a potent mitogen for a variety of cell types of mesodermal and neuroectodermal origin, including fibroblasts, endothelial cells, and melanocytes in vitro"
Thanks everyone for the nice comments, finnaly I have decided to use a TPO mimetic drug that seems to work in mice as well. Nevertheless, good to know about HST1 as well. Best.
Wei Zhong The drug is called Romiplostim. You have to inject 100 µg/kg two times before your experimentation with 3 days between injections. So for example Tuesday Injection 1 and Friday Injection 2, and you can perform your experiment the following monday. It worked perfect for me on Balbc mice. You can reach 5 times increase in platelet count. (Protocol courtesy INSERM U949 - Etablisment Français du Sang (EFS) Alsace, FRANCE)
