I am trying to clone Lentiviral expression vector, and it is suggested for NEB Stable E.coli strain by Addgene. can i use DH5 alpha or any other strain for this ?
If the DNA you are trying to clone has numerous repeated sequences, then using a string like Stable is appropriate. However if you don't have repeats, then you can most likely use any other strain you have available like DH5.
The 3' and 5' Ltr regions of the viral construct have a propensity to recombine so it is best to use a strain that will minimise this happening. Stbl3 from invitrogen are quite good as are NEB stables. The nature of the construct build also dictates cell choice. If you are using a gateway system like invitrogen's lentiviral kits then you need the ccbd gene containing cells. If using NEB's HiFI gibson assembly kit, I have had much greater succsess with NEB stables than STBL3. However I prefer the general DNA quality from STBL3 cells for viral production. You can clone in routine cloning cells like DH5 or Mach1Tr it may pay to reduce the temperature of incubation to around 30 degrees. However if you consider the time and effort and especially cost you are going to put into making the virus, purifying and titrating it I would always chose the strain that maximises my chance of succsess, for me it is Stbl3 and NEB stables, and for viral production in that order.
