I would like to prepare small unilamellar vesicles using probe sonicator. Therefore, I am intended to buy one probe sonicator. Can anyone tell me which sonicator to buy (Specification)?
For SUVs preparation, I have used a tip-ultrasonicator (Misonixs 3000, Qsonica, Newtown, CT operating at frequency 20 kHz). You can download the manual here with specifications. http://www.sonicator.com/literature/manuals/Sonicator3000Manual.pdf
The protocol for SUVs preparation can be found in Biochimica et Biophysica Acta (BBA) - Biomembranes Volume 1848, Issue 12, December 2015, Pages 3175–3180.
Any tip sonicator with power above 150W will work well to sonicate aqueous suspensions of lipids (formed by hydrating a lipid film) and transform it in a pale-blue transparant aqueous suspenstion containing SUVs. The concentration of 5-10mg/ml of lipids is ideal.
