I have collected several samples from tobacco filed, I'm looking for TSWV.
I need to purify TSWV from other viruses in my samples.

I afraid of mixed infection in my samples with TMV, TSV, TSWV and other common virus in tobacco.

Actually, TSWV is very common virus. İt can be difficult purify from other viruses on any plants. But , If you work about molecular diagnosis the virus, you can select a good primer for RT-PCR and PCR...Furthermore, you should work a good RNA extraction for PCR....
NO, there is no very specific host for TSWV. It has a wide range of host specificity.
You can use monoclonal antibody to detect the viruses separately and try to pick the host infected by single viral sp. (TSWV). If you once able to identify the host with mono culture infection, you can easily multiply your desired strain (TSWV) as a pure culture in healthy host for your downstream work.
RT-PCR and cloning will give you the pure culture that you can mechanical inoculated in the indicator plants which are suitable for TMV or ToMV.
The use of monoclonal is another method but even then you have to follow the molecular diagnostics to get the pure clones.
