I want to stain bull sperm cells (dead/alive) with Hoechst 33342 (10 mg/mL in H2O) and don't know how to do it properly. I will be grateful if you could help me. Best regards and stay healthy.
I have not specifically stained sperm cells but I have used Hoechst 33342 to stain cells often. You want to mix the lyophilized powder in ddH2O to make a 10mg/mL stock solution that can be stored in 4 degrees for up to 6 months or at -20 degrees for longer (several years). Dilute the Hoechst 33342 stock solution 1:2000 in PBS for staining. Incubate for 10 minutes protected from light, then wash and image the cells. You can also image in the staining solution if you are using a dish if you would like.
