I have read some protocols but they have some chemicals like borosilicate green which are not easily available in lab so I want such protocol that can be easily done in lab and dont take maximum time.
Conventional gravimetric method of analysis is available for estimation of total alkaloids where the plant material be defatted initially with hexane followed by extraction with ethanol or methanol. The concentrated extract can then by acidified with 10% aqueous sulphuric or acetic acid. Allowed to stand and basified with ammonia solution and the basic alkaloids can then be extracted with 2 volumes of chloroform or dichloromethane. Evaporate the solvent and the residue weighed.
The percentage of alkaloid was
calculated using the following formula:
% = W/Y x 100
Where W = Weight of alkaloid content extracted and
Y = Weight of powdered plant material.
This is the preliminary wet lab method of estimating total alkaloids.
