I have a ex vivo drug screening system for leukaemia, but I realised some samples contains both leukaemia and a significant amount of healthy cells (5-30%). I am looking for a no-wash (or better no-touch) protocol to stain the cells for leukaemia-specific cell surface marker (e.g. CD123 if the patient is tested CD123+) to streamline the experiment, and complement with the current protocol using CyQuant Direct. Has anyone had experience on this?
My initial thought is either just dump some antibodies in it and hope the background is not high, or use pHrodo to tag my antibody and hope it gets internalised on binding. There are some articles (e.g. Article Real-Time Imaging of Cell-Surface Proteins with Antibody-Bas...
) that provides an ideal solution, but I doubt that I can mass-produce this type of antibody at the moment even if it works perfectly on most antibodies.
