I try to stain cell to evaluate the cell differentiation by immunofluoresent assay.
Our hydrogel is temperature sensitive gel, which turns to solid form in 37 'C.
In fact, I tried to do cryosection of hydrogel and stained cells by immunofluoresent assay to evaluate effect of cell differentiation, but there were some problems (gels were easily destroyed).
As our results, live and dead staining was well done in 3D hydrogel form. So I think it is possible to stain the 3D hydrogel itself, but I'm not sure it can be possible.
Is there anyone to do staining of cells encapsulated hydrogel itself without cryosection?
