I have compared the water content of mutant and wild-type plants by weighting them before and after dehydration, however, what I really want to look at is the real-time, in vivo water content in their vascular systems. Any suggestions?
Check the ultrasonic resonance method. It is not invasive and not destructive. In Vitis vinifera, for example, allows for continous estimation of turgor changes.
