There are various protocols for the determination of plant hormones. the one which i use is solvent extraction followed by HPLC. since auxins and gibberellins and ABA are all acids, then they could be determined in the same sample following the extraction.they are usually extracted using 80% methanol for 48 hours in darkness at 4C. the n the extract are evaporate to aqueous phase at 35C. the this phase is made to 50ml with distilled water. make the pH 2.5 and extract with freshly distilled di ethyl ether three times., and evaporate under vacuum to 2ml. then perform HPLC using internal standard for IAA, GA3 and ABA. for cytokinins, make the pH of the aqueous phase 8.5, and extract with ether as above. use zeatin as internal standard for cytokinins.
