I've been recently using GE Sepharose® CL-6B gel (an size-exclusion gel) to separate polysaccharide samples. But I accidentally pumped air into the column and dried out the well-packed gel inside for about 8 hours. I'm wondering can this "drying" process cause irreversible damage to my gel ?
True drying can irreversibly damage the gel. I do not think that happened with your gel. Anyway rehydrate the system and test it.
I do not have experience with your specific coloumn.
But in the past i experienced a similar situation with a pre-packed 26/60 supedex 75 coloumn connected to an old FPLC which went dry during the equilibration O/N due to an error in the programm . The coloumn remained dry O/N but after long reydratation (about 10 CV of coloumn) the coloumn was able to perform well.
It depend if you will be able to completelly remove the air from the coloumn and if it affect the resin compactness.
However as Omar suggested, the only way to know it is perform a long reidratation and test it with a standard sample.

I did use the rehydrated gel before & it did work & gave very good result,using HPLC is excellent but it is not available in each laboratory and department ..its columns are very expensive .So we are talking about how you can carry on with what is available to most.I will very grateful to you if you could supply us with the columns & your developed resins.
