I am working with human adipocytes. The differentiated adipocytes are full of lipids and therefore float in standard cell culture medium. I think that decreasing density of medium would prevent floating of adipocytes resulting in culture conditions more similar to that in body. However, i do not know if this is doable by any mean.
Have you considered adding a suitable detergent to your solvent? There are several detergents available for molecular biology work.
Thank you for your advice Vicente. I have not consider using detergents since I would like these cells to survive. Maybe I am mistaken but I thought that concentration of any detergent which would stop that floating would also results in cell lysis.
@PeterLenart I am very sorry I misunderstood your question. I thought you were working with lipids in solution instead of adipocytes in liquid media. FYI a Japanese team lead by H. Sugihara developed a "ceiling" culture and also a 3D collagen gel culture to get around the problem of adipocytes floating on liquid media because of their high buoyancy (low density). These culture methods are probably what you need. HTH. Sorry again for the confusion.
