Actually I want to check toxicity studies of one human dose of a drug given to Wistar rats. Should I use the same human dose or as per the rule of FDA formula, convert that human dose into a rat dose? Because the animal (rat) dose should be 100 times higher than the human dose.

To convert human dose in mg/kg into rat dose in mg/kg, multiply by 6.2. For details, check Table 1 in the following guideline: 'Estimating the Maximum Safe Starting Dose in Initial Clinical Trials for Therapeutics in Adult Healthy Volunteers'.
