I am trying to isolate Bacillus amyloliquefaciens from plant rhizospheric soil. I have a few isolates that are starch hydrolysis positive. I am assuming they are bacillus spp. based on colony morphology and a few biochemical test (Oxidase, catalase, VP) results but I could not determine whether they are B. amyloliquefaciens or not. So some suggestions about confirmatory procedure (or test) of B. amyloliquefaciens will be appreciated. Thanks in advance.
DNA sequencing of ribosomal RNA genes should allow you to tell if the isolates are B. amyloliquefaciens.
You might also want to look at lactose and gluconate fermentation:
