I'm doing a lot of experiments with sperm cells and I want to start quantifying their motility to see if it is impacted by any of my experimental conditions. I could do this myself just by counting motile vs immotile cells on the videos I've captured, however I will be doing a lot of this and it would save me huge amounts of time if a computer program could do this for me or at least help in some way. I know ImageJ/Fiji are able to count cells but as far as I'm aware they can't quantify motility in a video.
If anyone knows of any software or other ways to do this that would be really helpful (hopefully free, but if I have to pay for some software I think it would be worth the investment). Thanks in advance for any help!

Thanks for the response - how exactly would I go about doing this in ImageJ? I don't need to calculate their velocity, I simply want to know how many of my cells are moving and how many are not - your method looks like it could be of use to me though.

although I think José Luis asnwer is right to do it and to meassure the Velocity, there might be a faster way in using stack functions.
i.e. You could calculate the total number of cells from a single Frame.
Than you could make use of the "Delta f down" or "Delta f up" function to subtract our current Frame from the nex one. so that only differences (moved cells) show up. You can than count their number as well. done :-)
You might want to play a little bit with the temporal sampling rate of your stack i.e. deinterleave some slices (i.e. 10) to get a better result (speration) between moved cells and non moved onces.
