Hitrap Heparin HP is coupled to the Sepharose High Performance base matrix via the N-hydroxysuccinamide coupling method and in case of Heparin sepharose 6 FF, Heparin is linked to the Sepharose matrix by reductive amination. What is the difference?
N-hydroxysuccinimide reacts with primary amines (N-unsubstituted glucosamines) of Heparin. However, in Heparin the amount of N-unsubstituted glucosamines is rather low (depending from the source/supplier).
Using reductive amination you can introduce an additional amino group at the reducing end of Heparin for coupling reactions.
If they use for both products the same Sepharose, which contains an amine-reactive chemical group (N-hydroxysuccinimide), there should be no great chemical difference.
