I am working in quantifying the bacterial metabolome. I want to lysis the bacterial pellet for extracting the metabolites. Is there any established protocol to lysis the bacterial cell pellet?
Will addition of water cause osmotic shock in gram positive organisms?.

For genomic DNA extraction from both Gram + and - bacteria, I use 5 ml of TES buffer (5 mM EDTA, 50 mM NaCl, 30 mM Tris pH 8.0) added with 1 ml of lysozyme (10 mg/ml) to resuspend the pellet and cause the lysis of the bacterial wall in the very first step of the protocol. If you increase the concentration of lysozyme, you can obtain a higher yield from Gram +, I guess.
