I'm culturing CHO cell using media of SAFC bioscience.
When culturing at 20 ℃ , there is no pellet.
However, when culturing at 37 ℃, the brown pellet come out after centrifuge.
Is there any suggestion that what kind of media component makes those pellet?
Or is there any suggestion for solvent for dissolving that pellet? (for LC-MS analysis of pellet)
It would help to know which medium you are using and what CO2 concentration in the incubator. Sometimes salts like phosphate can precipitate out of solution at the wrong pH and sometimes it can be fungal or mycoplasma infections. Have you looked at the precipitate under a microscope to see if it is crystalline or looks like cells?
Use a different Bottle of medium anyway
The precipitation from media could arise sometimes from FBS component. If you are preparing culture media yourself from commercially available powder, there is a chance that residual phosphates can result in precipitation (not necessarily brown). You could spread the brown pellets on a microscopic slide and see for what it is.
Another possibility could be bacterial or fungal contamination.
Regarding assessing the pellet by LC-MS you can extract the pellet with either acetonitrile or methnol for assessment.
