tRNA derived fragments are a novel regulatory ncRNAs with distinct biological functions in cancers and stress-induced diseases. Our research group has worked on tRFs for a while. The major method we used was transient transfection of a synthetic tRF mimic or an inhibitor to cells, similar to the methods applied for miRNA research. Yet, we occasionally encountered the transfection efficiency problem and the bench effect problem, which is really costy and time consuming. And this made me wonder if there is an expression plasmid system for tRNA derived fragment, similar to the plasmid for shRNAs or miRNAs expression. Maybe because of my carelessness, I have not found any info regarding this expression system from pubmed yet.
