I would like to know if there is any in silico method to predict a suitable chromogen which is to be bound to a antibody in a specific antigen-antibody reaction?
Hi Vaisakh, I don't know what your specific assay is, but most antibodies have only 3 reactive groups that can be used to attach a reporter group: -NH2, -COOH and carbohydrates.
The method is important. However to have a polivalent antibody suitable for ELISA, Western-blot and even flow cytometry biotinilation is a good option. Biotinilate is very simple. There are commercial kits to do it and it is very easy to biotinilate (I generally prefer sulfo-NHS-biotin). Then you can use as "secondary antibody" streptavidin labelled with different chromogens depending of the technique you are going to use.
