I would like to measure the increase in sporulation of a filamentous fungus on a solid culture. This fungus does not grow well in liquid culture so I need a method that will work with petri plates. I could make several replicate plates and measure the sporulation of each at a different time point, but I think this could be open to many sources of error. Are there any other suggestions?

Broth culture is still better if you need to measure sporulation. If the fungi does not grow or sporulate well:
1) Check if your fungi produce spore or not (Some fungi especially Basidiomycetes do not produce spores in plate or broth but you can get the spores from their fruiting body). If your fungi produce spores on plate, then it should not have any problem growing and producing spores in broth.
2) You can modify the growing media to boost spore production, such as adding 1% yeast extract in the media.
On one time, you can take small agar discs (10 disc for example) with a punch, in the diagonal of the culture , from the center to the periphery.On the other time made the same procedure, to other diagonal.
