I am trying to create a construct consisting of the GnRH promoter region from a chicken and the SV40 large t-antigen (TAg) encoding region for my master's thesis (to later create an immortalized avian GnRH secreting cell line). I was having trouble trying to isolate the TAg and ran across one that is already ligated into a plasmid but I am unsure of what regions of the TAg it includes. Has anybody ever heard of this? Thanks!
Rachael, I think Addgene (http://www.addgene.org/) is your best bet. It looks like they charge $65 and you can browse for the exact vector you need (they have a ton that encode the large TAg). I would recommend ordering one that has complete plasmid sequence available, a referenced publication in which it was used, and a complete vector feature map.
