My sample has both vitamin A palmitate and vitamin E (2%). Does vitamin E interfere in the assay of vitamin A palmitate by spectrophotometric method?
Is there any reliable method to estimate vitamin A palmitate specifically by UV-Visible spectrophotometry (NOT HPLC)?

Since I don't know the details of your sample, it is not feasible to give you a clear answer, but I think it would be tough if you just use the UV spectrum.
Although it is likely that the absorption maxima of tocopherols and retinyl palmitate are not close to each other, if the concentration of one of them is extremely high, it will affect the absorbance of the other. Also, in the case of biological samples, the matrix will affect the results.
I think you should measure the absorbance at the maximum absorption of one or the other and compare it with the result of mixing the two.
You could then check their accuracy, repeatability, and intermediate precision.
