Is there a comprehensive (perhaps proteomics-based) approach that will screen out autoantibodies from blood and also (reliably) predict their target? I am interested in autoantibodies against CNS antigens. Any other simpler method not involving omics is also welcome.
The most traditional method to detect autoantibodies is indirect immunoflurescence.
To do this, one must have the needed tissues (biopsies or surgical pieces) from animals or humans according to the different targets. Cut the biopsies in 5 micrometer sections on microscopy slides, cover them with the serum sample to be tested diluted 1/10 in PBS buffer, incubate 20 minues, wash the slides in PBS 20 minutes, cover the sections with anti- human gamma globulins conjugated with fluoresceine (commercially easily available)(if the sample is from humans of course, otherwise anti gamma versus the appropriate animal) incubate 20 minutes, wash the slides in PBS 20 minutes, Mount the slides with a coverslip and read at the flurescent microscope.
There are plenty of publications on this method.
Alternatively to immunofluorescence, there are ELISA kits working very well. Two Companies selling these kits are DIAPRO Diagnostic Bioprobes, MIlan Italy and Euroimmun Germany, which has also a subsidiary in Madrid, Spain. Both Companies can be explored on the internet web.
