I'm guessing a phloem-specific stain for grasses would work as well. I have a few barley lines carrying a phloem-specific reporter (DsRED), but I want to make sure the expression is indeed coming from the phloem. It would be great if there was a stain that distinguished the phloem from the xylem. I've been combing the literature, but haven’t found anything yet, so I thought I'd ask you folks.

We used to use aniline blue to stain phloem. Have you considered this option before?

Try a double staining, safranin-fast green, you will see clearly the xylem stained red and phloem dyed teal.
