I am doing a research on lignin and cellulose degrading bacteria. Upon isolation and DNA sequencing of the lignolytic and cellulolytic bacteria, FTIR, Raman, Atomic Force Microscopy, SEM, SEM-EDAX was done for analyzing topological changes due to degradation (other conventional enzymatic and microbiological analysis was performed). Then the best performing lignolytic and cellulolytic bacteria were inoculated into rice straw and the topological changes in the substrate were analyzed using FTIR, Raman, SEM, SEM-EDAX, XPS, XRD?
Since I am interested in topological changes due to bacterial degradation in lignin, cellulose and rice straw, is there a need for doing GC-MS?
We plan on doing GC-MS for pathway elucidation in the next part of the research.
