I was trying to take my DNA out of solution to resuspend it in a new buffer. I placed my samples in the SpeedVac and evaporated off the buffer until a white solid remained. I am concerned that this solid contains some of the salt from the buffer. Does anyone know a method to correct what I have done?
Unless the DNA started in a volatile buffer (ammnoium acetate or ammonium bicarbonate), when you evaporated all the water, the buffer and any salts that were in it were left behind, which is the white solid. Just add the water back to the solid to the original volume to get back to where you started. You can change the DNA into another buffer by dialysis against that buffer, as Sebastian suggested, or you can use standard DNA precipitation methods (e.g. salt and ethanol) to pellet the DNA, then resuspend the pellet in the desired buffer.
