Back in 2002 or thereabouts, some Japanese researchers postulated the existence of a Diels Alderase enzyme that could carry out 4+2 cycloaddition reactions. I have seen some theoretical papers in the past year but I am unclear if a definitive enzyme has been isolated and characterised. I would new interested to know what the current status is.

They have structure/biochemistry for this one. Paul Race did the structure, Jem Stach isolated the enzyme and did some of the biochem.

Bryan - I take it you read Craig's article ( Townsend, C. A. (2011), A “Diels-Alderase” at Last. ChemBioChem, 12: 2267–2269). There are more recent refs ..but I'm out of that loop now. The only work I've done on this was (Ligulaverin A, a Structurally Novel Sesquiterpenoid-derived Metabolite Possibly Formed via a Biological Diels-Alder Reaction; Y. Zhao, S. Parsons, R.L.
