We want to quantify LPS (endotoxin) levels in human plasma. There are several assays from Lonza, of which QCL-1000 (endpoint, chromogenic, based on Limulus amoebocyte lysate) is the most amenable to our laboratory's capacities and needs. Have you had problems with this assay? Are there any other assays from other manufacturers?
There have been some concerns raised about the sensitivity of this assay for some time. Various groups have had reproducibility issues with the assay. The manufacturer even recommends against using the assay for reliable detection in human/animal samples. I would say if you're going to use this or any limulus amoebocyte based assay you need to be very diligent and use all of the recommended controls/standards plus a spiked serum standard in a negative control background. If you wanted to increase the confidence in your results you could pair it with a simple 16s rRNA pcr assay (taqman has one) or even a sCD14 ELISA.
Hi, Kai. I'm actually looking for advice. From what Joshua says, I might go for 16s rRNA PCR, unless I find the way of reading the chemoluminiscent rFC assay by Lonza, that seems to be free of several downsides of the amoebocyte lisate tests.
