Based on Promega's manual, I have tried to select colonies of JM109 (Promega Ref. No. P9751 - non competent) on M9-B1 minimal plates for processing to prepare competent cells. I have got the tiny colonies that look like pinpricks. Is that normal and shall I proceed with those colonies or what modifications do you suggest? I have attached the relevant information of protocol and product details.
