I would say using THP-1 as representative of microglia is far from optimal. Although they might share similar functions and surface markers, they have different origins (hematopoietic stem cells versus yolk sac progenitors) and microglia is far more plastic (morphologically and functionally) compared to the THP-1 macrophages. If you are studying human microglia, it is probably important to have a good model that reflects all the functions microglia can do, beyond immune response.

why do you choose THP1 cell line? how will you differentiate this cell line towards microglia cells ? be careful...

thanks for your explanations, actually I'm kind of agree with you and I think that probably I can't use differentiated THP-1s as microglias.

I think that THP-1s are suitable for my study, because based on similar works , THP-1s are one of the promising choices for studying human monocytes/macrophages manner in vitro. I'm using PMA ( Phorbol 12-Myristate 13-Acetate) for differentiation of THP-1 into macrophages.
