Slide chambers have limited space for a staining procedure and are often covered up by on or two sectioned samples.
Because of this issue I was wondering if I could stain first and than section the samples. (Would save time and reagents)
I am interested in chicken heart and it is dissected from the embryo, thus the staining solution is able to reach the inside and outside the heart. Just in case this changes the matter a bit in regard to accessibility and tissue thickness.
You can to stain Hematoxylin and Eosin after to sectioning of tissue.

IF YOUR ARE USING PARAFFIN BLOCK MEANS YOU HAVE TO REMOVE THE WAX THEN YOU CAN STAIN IT ........ CHICKEN HEART PLEASE TELL THE PROCEDURE HOW YOU ARE FIXING THE TISSUE ARE YOU USING 10% FORMALDEHYDE......
I am fixing the whole embryo in 4%PFA overnight at 4°C (~16h) and dehydrate next day stage wise with Methanol and leave them at least 1 day in Methanol at -20°C. Than i stage wise rehydrate until the stop floating. I dissect the heart out using forceps and than I usually move over to 20% sucrose EDTA overnight at 4°C before I embed them next day in Gelatin in phosphate buffer.
I wanted to do the staining before embedding but i think I tried once with Kernechtrot (nuclear fast red) and the staining disappeared in heated gelatin at 37°C.
Anyway there doesn't seem to be a short cut for staining before sectioning.
