I want to mix and solubilize my compounds that are stored in Assaybuffer with 2% DMSO. Monoshaker doesn't seem to work efficiently even at 2000 rpm and mixing by hand is not an option.
So are your compounds already dissolved but are stored frozen? If so then sonicating probably won't help. If you are dissolving compounds it will help break the particles up but you'll have a concentration gradient (high at the bottom). I see you're from Evotec, we've been using 384-well cell carrier plates in our OPERA to screen yeast libraries and mixing on the shaker works well, but then we have only a 30 - 50 uL volume. Is your volume close to the maximum for the plates? If so it might be possible to seal the plate with aluminium foil (or something similar), shake very, very hard then spin the solution to the bottom of the plate in a centrifuge with appropriate "buckets".
Thanks for your answer James! Actually I am working with SPR. The 384 Deep Well Plates we use have volumes of 100-200 µl in them. I guess your approach with the sealing and shaking could work, i will try it out and give some Feedback.

Another thought, are you using round or square wells? Round wells will allow a greater vortex to be created and with less turbulence there will be less splashing, furthermore if you pulse the shaker rather than have it on continuously it will mix better. Funny how the simple things can be the biggect stumbling block, good luck.
I find the Eppendorf MixMate to be a very good instrument for mixing in 384-well plates.
Thanks Adam! I had a look at the MixMate it seems well suited for what I intent. Do you have any expierience with 384-Deep-Well Plates using the MixMate? In the description it only named 384-well plates with low Volume samples.
As long as the deep-well plates have the same footprint as the mixmate will work fine. That's what we use also. It's very stable in the horizontal plane so the deepwell plates shouldn't be a problem.
I haven't used the MixMate for 384-deep-well plates. It's been able to mix everything else I've tried.
