I've read in other questions that some cultures of Chlorella are preserved in agar plates and in refrigerator. But I was wondering if there is need of light also in agar plates?
I am culturing a marine diatom Phaeodactylum tricornutum also in agar plates, but I've left them at room temperature and near window for solar natural light. I've also been told that this strain can not be stored in -20°C refrigerator, so I was wondering also if it is a good idea to store it in 4°C refrigerator. Which are your experiences about it?

You can first allow the cells to grow on agar plates as per your protocol. Once the cells are ready, you can store the agar plates at 4 deg C. Light is not required for storage.
Yes first there are some methods for preservation you can preserve algae by using logal solution if you grow algae in broth media .. formalin also can be used ..... Freezing is suitable for algae grow on broth you don't need light for preservation and there is some reserved food inside algae
But take care some algae will die by freezing
Hello, refrigerator light should not affect the strain, as it does not produce a lot of UV (ultraviolet), so I would not care too much about that. The growth of the cells should be at room temperature, in their natural conditions. Then you can store them at 4 deg. -20 C will freeze them and probably cause crystal formation inside the cells, which then will damage them irreversibly. Best regards.

You can preserve your desired algal samples on agar plate in the refrigerator at 4 deg. C for little long time (1-3 years) Or more by the proper cover with parafilm tape then kept it into a plastic box and preserve it into refrigerator.
You can also preserve your sample at -20 deg. C and -80 deg. C with DMSO Or glycerol and liquid nitrogen that avoid the crystal formation, you can preserve your sample through this method for very long time (10 y.) Or more. I have attached the file of DMSO protocol.

Hello, refrigerator light is not needed. i have preserved Chlorella sp. at 4 d.c. using agar plate made of 5% strength of LB plus BG11 nutrient media. this formulation is necessary to be exact but in my case it works well.
