Hi Bakul, you can check bacterial cell viability through different staining methods, for example dead cell can be stained with trypan blue.

Many kinds of literature exist in which MTT is used to check bacterial viability (live cell). The method is more or less similar to eukaryotic/mammalian cell culture. I am sharing the papers you can go through for more information.

I had though about staining with Trypan Blue for measuring viability but it would be a very cumbersome process because I have to check a large number of samples simultaneously. This is what prompted me to consider MTT as an alternative to cfu counting. I read a few papers on MTT assay with bacterial cultures, all of them suggesting different protocols that confused me about its feasibility.

@Bakul piplani did you finally used MTT for cell viability of E.coli? And did it work ? I also need an alternative to CFU and I'm not sure if i can use this method
