Currently I am looking forward to start with the standarization procedures to isolate T cells using the Dynabeads® Regulatory CD4+CD25+ T Cell Kit. The problem is that in the lab we have the MPC™-1 magnet, but the manual clearly states that this magnet should not be used.
I recently ordered the Dynamag magnet that is compatible with the kit, but in the meantime I would like to advance. I want to probe that the MPC™-1 magnet is really useless and also I will try with the
PureProteome Magnetic Stand form Millipore
Before going any further I want to make sure that this attempts will provide results, so that it will be very useful for me if you guys let me know your experiences regarding the use of magnets different from those recommended by any kit, specially if someone has used a different magnet from the Dynamag with the Dynabeads® Regulatory CD4+CD25+ T Cell Kit .

I have used magnets interchangeably although not the ones you mentioned. If the manual clearly states that MPC™-1 magnet should not be used, I would not. There may be some incompatibility issue there. You may have better luck with the Millipore magnet. However, purity of the isolated population may be affected when using alternate magnets. The frequencies of regulatory T cells are low and so a small difference in purity may have consequences in your downstream applications. The best approach would be to call ThermoFisher and ask them whether you can use magnets interchangeably and if so, which ones are compatible with the kit.
Hi, all DynaMag magnets can be used except for the MPC-1 for cell isolation. The challenge is the negative isolation part of this kit which uses smaller MyOne beads (1µm). These beads require a stronger magnet than the MPC-1. But all other DynaMags work really well.
