I am planning to perform an experiment in mice to check effect of tolerance inducing agent for factor 8. Since I do not have Hemophilia a mice, I was thinking of using normal mice. As I believe, there is 20-30% difference in mouse and human factor 8 so when injected with adjuvant, recombinant human factor 8 should induce anti factor 8 antibody and other immune responses in mice. However, I see its long time people have stopped using normal mice and they are using only hemophilia mice and I don't see any reference referring to this (immunization of normal mice with hfactor 8).
I will appreciate if some one share their experience here, if they have one.

One quick information for you, if you are trying to induce anti-factor8 antibodies, you need to inject the recombinant protein with an adjuvant to promote antibody development, and the best one is complete Freund's adjuvant. But CFA if toxic, and most animal research restrict it's use.
Second, just because you get anti-factor8 antibodies, does not mean you will induce hemophilia in the normal mice. Hence the use of the hemophilia mouse strain. Much more consistent disease presentation.
If possible, I would suggest you try to get a breeding pair of the hemophilia mouse strain and generate a colony on site. The reason is if you use a model that is NOT in line with what is currently used in the field, you need a LOT of strong data just to prove that you generated an acceptable model of the disease.

Yes, I agree with you and I am planning to get hemophilia mice. Mean while I was thinking of doing some experiment with normal mice just to have some hints on my hypothesis.
